中文摘要 分離自猴耳草(Peperomia dindygulensis)之內生真菌菌株PDL-005,具有產生抑菌效果揮發性氣體之特性。該菌株可抑制柑橘綠黴病菌生長與殺菌之效果,透過形態學特徵與ITS序列,被鑑定為Nodulisporium sp.。進一步以各種不同培養基質,測試對PDL-005菌株生長與產氣效果,結果指出PDL-005菌株培養蔗渣後,可透導該菌株產生更有效抑制病原菌之氣體的特性。以GC-MS分析有效抑制病原菌之氣體成份,結果得知2-ethyl-2-hexenal 2,4-dimethyl-1,3-cyclopentanedionePDL-005菌株所產生揮發氣體中具抑菌成份的主要物質。於實際防治柑橘綠黴病之試驗中,證實以蔗渣培養PDL-005菌株後,可降低柑橘綠黴病的發生達100.0%。即PDL-005具作為防治貯藏病害柑橘綠黴病之生物製劑的潛力。
英文摘要 A volatile-producing endophytic fungus, PDL-005, with bio-fumigation activity was isolated from leaves of the medicinal plant Peperomia dindygulensis. PDL-005 showed an inhibitory efficacy or fungicidal ability against Penicillium digitatum Cmep1-3. The PDL-005 endophyte was identified as Nodulisporium sp. based on the morphology and ITS nucleotides. Testing different ingredients revealed that bagasse was the best material to produce active compounds, which exhibited good efficacy against the test pathogen. GC-MS analysis revealed that 2-ethyl-2-hexenal and 2,4-dimethyl-1,3-cyclopentanedione is the primary compound contributing to the inhibitory capability on radial growth of the pathogen. Moreover, PDL-005 cultured in bagasse reduced green mold incidences on orange markedly, sometimes up to a 100.0% reduction. Thus, PDL-005 could be potentially used as a bioagent to control postharvest disease caused by P. digitatum.
發表成果與本中心研究主題相關性 柑橘綠黴病為柑橘類作物之重要貯藏病害,目前的管理模式皆以化學藥劑處理,而化學藥劑處理後對農業環境之永續經營有極大的副作用。本研究分離具產生抑制病原菌之揮發氣體之植物內生性菌株,並開發可促進其產氣之基質,且具有防治柑橘綠黴病之潛力,未來可開發成防治作物貯藏病害之生物製劑,與中心研究具相關性。
